Enteropathogenic Escherichia coli (EPEC) frequently induce diarrhea in human infants and in young food producing
animals by causing attaching effacing (AE) lesions of the intestinal epithelium. Their pathogenicity has been proven for rabbits, mice, lambs, and for colostrum deprived newborn pigs but not for conventional weaned pigs. Here we aimed to compare in vivo pathogenetic potential and virulence genotypes of intestinal (n=6) and faecal (n=4) isolates of EPEC from weaned pigs. For this purpose we inoculated ligated ileal loops of 4 weeks old weaned pigs for assessment of EPEC adherence to ileal enterocytes by histology and immunohistology. We also determined the virulence gene patterns of EPEC using an E. coli PCR-microarray. The intestinal EPEC isolates of sero-/intimin types O45:H11:eae-β, O49:NM:eae-β:, O84:H7:eae-γ, and O123:H11:eae-β formed adherent microcolonies of EPEC on ileal villi more frequently (p<0.05) than faecal isolates of O28:H28:eae-NT, O108:H9:eae-β, O145:H28:eae-γ and O157:H2:eae-β. Microarray and PCR analysis of both groups detected numerous virulence genes characteristic to LEE (locus of enterocyte effacement), and non-LEE pathogenicity island of EPEC and enterohemorrhagic E. coli (EHEC). Overall, intestinal isolates carried more (p<0.02) virulence genes than faecal isolates. Five of 6 intestinal isolates contained the genes efal, lpfA, and tsh contributing to mucosal adhesion in contrast to the faecal isolates with none of these genes. Thus, the ileal loop model in weaned pigs is suitable to differentiate between intestinal and faecal isolates of porcine EPEC. Diverging virulence of the intestinal and faecal isolates is suggesting dynamic evolutionary events within the population of porcine EPEC. |
